Effect of extender and removal of seminal plasma on quality of frozen Beetal buck semen

Abstract

A total of 20 pooled ejaculates comprising 84 ejaculates collected from five adult Beetal bucks maintained at Goat Research Station, AAU, Burnihat were used in study. The pooled ejaculates were split into two equal parts and one part of pooled ejaculate was split into three parts and subjected to washing. Both washed and unwashed parts were extended in three extenders viz, Tris extender containing 20 percent egg yolk or 1.5% soya lecithin and commercial Optixcell extender. The extended semen was filled in 0.25 ml French straws and frozen and stored in liquid nitrogen. The frozen semen was evaluated on following day for sperm motility, live sperm, intact acrosome and HOST reacted sperm. The mean post thaw sperm motility in unwashed and washed semen in Tris extender containing 20 percent egg yolk or 1.5 percent soya lecithin and in commercial Optixcell extender was 57.20±0.97 and 62.05±0.76, 51.05±1.06 and 54.10±1.04, and 59.65±1.17 and 61.95±1.13 percent respectively. Corresponding values for live sperm were 63.29±1.03 and 66.40±0.94, 53.67±1.27 and 55.67±1.34, and 62.44±1.00 and 64.86±0.88 percent; for intact acrosome were 60.77±0.82 and 62.18±0.86, 52.09±0.68 and 54.24±0.87, and 59.98±0.78 and 61.28±0.77 percent; for HOST reacted sperm were 48.70±0.82 and 51.35±0.90, 47.80±0.45 and 49.55±0.98, and 50.06±0.68 and 52.24±0.84 percent. In both washed and unwashed semen post-thaw sperm motility, live sperm and intact acrosome in Tris extender with 20 percent egg yolk and commercial Optixcell extender were significantly higher than Tris extender containing 1.5 percent soya lecithin.
