Genetic diversity of charcoal rot fungus Macrophomina phaseolina, using SSR markers

Abstract

The charcoal rot of soybean is one of important disease it cause 70% loss in soybean yield. The present investigation where an attempt had been made regarding diversity within this pathogen will help the development and release of disease-resistant soybean cultivar and contribute in understanding the population of biology of this organism. The genetic diversity was studied among the ten isolates using 6 SSR primers of MB series and SSR series. Six primers produced 72 bands. Out of 72 bands, 66 bands were polymorphic and average level of polymorphism was 91.66%. In the dendrogram, MP-1 (Akola) was found to have higher value of similarity coefficient (0.857) whereas MP-3 Amravati was found to have lower value of similarity coefficient (0.128). The data regarding the screening fourteen genotype of which four genotype viz., AMS MB 5-19, AMS MB 5-18, AMS-1001, AMS-77 were resistant. Four genotype viz., AMS-353, AMS-358, BRAGG, JS-335 showed moderately resistant reaction. Three genotype showed moderately susceptible reaction viz., AMS-243, JS-93-05, AMS 99-33. Three genotypes were highly susceptible viz., AMS 38-24, AMS-475, TAMS-38.
