Polymorphism in the minimal promoter region of elongation of very long chain fatty acids protein 2 (ELOVL2) gene and its effect on serum triglycerides in layer chicken
Abstract
Polyunsaturated fatty acids are important for human health. In extension of long chain fatty acids the enzyme elongation of very long chain fatty acids protein 2 (ELOVL2) play a pivotal role. The purpose of this study was to determine the polymorphism in the ELOVL2 gene's promoter region and how it related to the serum triglycerides in control layer chicken maintained at ICAR-Directorate of Poultry Research, Hyderabad. Using the conventional phenol-chloroform procedure, genomic DNA was extracted from blood drawn from the wing vein of 150 birds. Using gene specific primers, the promoter region was amplified by polymerase chain reaction (PCR) and the PCR product was then exposed to single strand conformational polymorphism (SSCP) to screen for polymorphism in the ELOVL2 gene's promoter region. Eight haplotypes and eleven haplogroups were found in the ELOVL2 gene's promoter region. The most common haplotype was H5 (0.348), whereas the least common was H3 (0.007). According to an association analysis, there was a substantial correlation between the haplogroups and the level of serum triglycerides. Serum triglyceride levels were highest in birds with the H1H2 haplogroup (161.85 + 39.28 mg/dl) and lowest in birds with the H5H8 haplotype (111.77 + 5.82 mg/dl). In summary, the findings showed that the ELOVL2 gene's promoter region was polymorphic and that it affected serum triglycerides.
