Molecular isolation and characterization of bioluminescent Photobacterium leiognathi from marine squid
Abstract
Bioluminescent bacteria play a crucial role in marine ecosystems, often forming symbiotic associations with marine organisms. In the present study, Photobacterium leiognathi, a bioluminescent bacterium, was isolated and identified from marine squid. A total of 30 samples were collected, and initial screening revealed nine non-luminescent isolates alongside a single luminescent bacterium. Biochemical analysis confirmed the identification of the bioluminescent bacterium as Photobacterium leiognathi. The isolated strain was further subjected to genomic DNA extraction and restriction site analysis to gain insights into its genetic composition and diversity. Genomic DNA was isolated using standard molecular biology protocols, and restriction enzyme digestion patterns were analyzed. The restriction site analysis provided valuable data on the genomic structure of the isolate, contributing to the understanding of its bioluminescence mechanism. The results of this study highlight the significance of isolating and characterizing bioluminescent bacteria for their ecological, evolutionary, and potential biotechnological applications. This research provides a framework for further molecular studies and potential utilization of Photobacterium leiognathi in various fields, including bioengineering and marine biology.
