Standardization of in vitro rooting and ex vitro hardening for in vitro propagation of potato (Solanum tuberosum L.) variety Kufri Surya
Abstract
The present investigation entitled “Standardization of invitro rooting and ex vitro hardening for in vitro propagation of potato (Solanum tuberosum L.) variety Kufri Surya” was conducted at Plant Tissue Culture Laboratory, College of Horticulture, Dr Y.S.R Horticultural University, Andhra Pradesh in a Completely Randomized Design. Sprouts were pretreated and surface sterilized and these sprouts inoculated into culture media with different growth regulator concentration for culture establishment and transferred into shoot proliferation media with different growth regulator concentration. The following observations recorded after 21 days for number of shoots producing roots, rooting percent, number of roots per shoot, length of longest root and root colour. Results showed best rooting response was observed by using IBA than NAA. It can be concluded that the MS medium supplemented with 1 mg/L IBA was found to be most effective among the different media. The ex vitro hardening showed that plantlets of Kufri Surya variety survived better on the cocopeat + Pseudomonas florescence.
