The Ayurveda formulation “Amrutadi” attenuates the inflammation in Freund’s adjuvant induced arthritis in rat

Abstract

Rheumatoid arthritis (RA) is a systemic, autoimmune and inflammatory disorder of joints with unknown aetiology. RA is primarily characterized by cellular infiltration and proliferation of synovium leading to the progressive destruction of the synovial joints in a symmetrical fashion. Complete Freund's Adjuvant (CFA)-induced arthritis is one of the oldest established animal models for testing arthritis treatments. It mimics RA in human and associated clinical and histopathological changes are comparable to those seen in human form. In the present study the rheumatoid arthritis was induced in rats by using complete freund’s adjuvant (CFA) @ 1 mg/ml to evaluate efficacy of Amrutadi churna and its constituents Tinospora cordifolia, Zingiber officinale, Tribulus terrestries and Sphaeranthus indicus. The analysis of biomarker, viz., TNF-α and interleukin-1 showed significant improvement in treatment groups compared to non treated and standard group i.e. diclofenac sodium. The haematological parameters, serum biochemistry, paw thickness and body weight also showed good improvement as compared to non treated and standard group. The has shown good results as compared to individual constituents of the formulation. It also showed reduction in ALP, ALT and AST and had no effects on kidney. Therefore, it can be concluded that the better effect of Amrutadi by combining different herbal ingredients might have resulted due to additive action of phyto constituent used in composition.
