Comprehensive study on explant surface sterilisation of in vitro culture of bamboo species (Bambusa balcooa)

Abstract

The experiment was conducted during the year 2023 at Tissue Culture Laboratory, College of Agriculture, Nagpur to study the comprehensive performance of surface sterilisation for In vitro micropropagation of Bambusa balcooa. Nodal segments were used as an explant for micropropagation collected from AICRP on Agroforestry, Nagpur. Total eleven treatments were studied along with three replication under Completely Randomised Block (CRD) design and statistical analysis was done. Out of various sterilization treatments tested, T9 - 1% Tween 20→0.2% Mercuric chloride→70% Alcohol→1% Carbendazim→0.1% Streptomycin (5→10→5→10→15 min.) shown 77.67% of aseptic culture followed by treatment T11 - 1% Tween 20→0.1% Carbendazim→70% Alcohol→0.5% Mercuric chloride→0.1% Streptomycin (5→10→0.5→5→5 min.) and T4 - 0.2% HgCl2 (5 min.). However, the lowest response for surface sterilization was observed in control treatment T1 - Control (DDW washing) (20 min.) with 24.33%.
