Evaluation of hematological changes in the blood collected from donor cows stored in CPDA-1 blood bags: A case study

Abstract

The purpose of the current study is to assess the haematology of cattle blood kept in plastic bags that contain CPDA-1. Six mature female cows that appeared to be in good health provided 100 ml of whole blood, which was then placed in a bag with 14 mL of CPDA anticoagulant—an anticoagulant meant to be used for the long-term preservation and storage of human whole blood. For 42 days, the bags were kept between 2 °C and 6 °C. Various hematologic parameters, including hemoglobin concentration, packed cell volume (P.C.V. %), red blood cells, leukocytes, platelets, MCH, MCHC, and MCV, were examined at the following times: day 0 (immediately following collection), day 7 (7 days later), day 14 (14 days later), day 21 (21 days later), day 28 (28 days later), day 35 (35 days later), and day 42 (42 days later). TEC was steady up to Day 35 and abrupt increase in Hb concentrations from Day 35 to Day 42 attributable to haemolysis. PCV changed in percentage from Day 35 to Day 42 and there was a significant drop in MCH and MCHC from Day 7 to Day 42. The WBC counts fell considerably from Day 21 to 42 days. As the platelets began to decline on day Day 7, they were extremely unstable. As the requisite haematological characteristics remained consistent for up to 25-30 days at 4 °C, Donor blood is capable of being stored in CPDA-1 blood bags enabling blood transfusion during emergencies. There continues to be a scope for developing blood bank over veterinary purpose.
