Standardization of culture establishment and shoot proliferation for In vitro propagation of potato (Solanum tuberosum L.) variety Kufri Surya

Abstract

Potato (Solanum tuberosum L.) is a vital dicotyledonous crop and tuber, ranking as the fourth most refined food crop worldwide, following wheat, rice, and maize. It is propagated asexually using tubers, which are highly vulnerable to a variety of fungal and viral diseases. Sprouts proved to be the most effective explants for initiating culture, leading to the highest survival rates with minimal contamination and mortality. The present experiment was conducted for standardizing growth regulator concentration for In vitro culture establishment and shoot proliferation. The observations recorded at 30 days for number of sprouts responded, percent of shoot bud induction, number of days taken for shoot initiation, number of shoots per explant, shoot length and number of leaves per shoot in culture establishment. For in vitro shoot proliferation, observations recorded at 21 days for number of shoots multiplied per explant, length of the longest shoot, number of leaves per explant, leaf area, average shoot thickness. Results showed better response with increasing BAP concentration upto 3 mg/L but the response was reduced with further increase till 5 mg/L. It can be concluded that for standardization of culture establishment and shoot proliferation of potato cv. Kufri Surya the MS medium supplemented with 3 mg/L BAP and 5 mg/L Kinetin was found to be most effective among the different media. The interactive effect of both BAP and kinetin showed better response than BAP alone.
