Abstract

Covalent inhibitors have re-emerged as a powerful class of therapeutic agents in medicinal chemistry, offering advantages such as prolonged target engagement and high potency. By analyzing recent studies, we provide insights into the development of covalent inhibitors, highlighting successful examples and discussing the challenges and future directions in this field.
