Abstract
This study investigates the free radical scavenging activity and protective antioxidant effects of baicalein and luteolin against lipopolysaccharide (LPS)-induced injury in goat pulmonary arteries. DPPH radical and NO Nitric oxide scavenging assay were performed to determine the free radical scavenging effect of baicalein and luteolin flavonoids using spectrophotometer analysis. For organ culture, goat pulmonary arteries were isolated from goat lung obtained from slaughter house and rings prepared in MKHS solution. Rings were placed in 24 well plates containing DMEM (Dulbecco’s modified eagle medium) supplemented with 10% foetal bovine serum. As a treatment, Baicalein and luteolin were added at 200 µM each in each well followed by LPS was added at 50 μg/mL to induce injury in tissues. Further, the tissues were incubated at 37°C with 5% CO2 for 24 hours before being analyzed for oxidative stress parameters. The DPPH radical scavenging assay revealed that baicalein exhibited a higher DPPH free radical scavenging activity (84.3±3.22%) compared to luteolin (81.6±3.18%), with IC50 values of 7.48 µg/mL and 8.85 µg/mL, respectively. Both compounds displayed comparable nitric oxide (NO) radical scavenging activities, with baicalein at 80.3±4.38% and luteolin at 79.9±1.38%, and IC50 values of 9.73 µg/mL and 9.43 µg/mL, respectively. Baicalein and luteolin significantly increased the level reduced glutathione (GSH) compared to the LPS group alone. Both compounds significantly decreased malondialdehyde (MDA) level indicating reduced lipid peroxidation. These findings suggest that baicalein and luteolin effectively scavenge free radicals and exert protective antioxidant effects by enhancing GSH levels and reducing MDA production.
