Detection of mutation in the BRACA2 gene for early diagnosis of canine mammary tumour

Abstract

The identification and analysis of different markers associated with tumour is very important to understand the cause of tumour and its early diagnosis in all species of animal including canine. Identification of mutations in BRCA2 genes by PCR-RFLP is an important technique for early diagnosis of canine mammary tumour. In the present study PCR RFLP of BRCA2 (Exon 27) gene was done for detection of mutation in canine mammary tumour and histopathology was done to study the change of tissue architecture in positive cases. Blood samples were collected in vacutainer tube with anticoagulant from 20 female dogs brought for treatment of breast cancer in College of Veterinary Science, AAU, Khanapara. DNA was isolated from blood and amplified BRCA2 (Exon 27) gene using specific primer. One half of the purified PCR product was used for restriction digestion and the other half was used for sequencing.There was a change of adenine to cytosine in the cancer positive samples at position 392 which caused the creation of a new restriction site for SsiI. Present study indicated that PCR -RFLP of BRCA2 (Exon 27) gene can be used for detection of mutation in canine mammary tumour.
